Cloning and Characterization of the Lysozyme Gene From Parapenaeopsis Hardwickii

  • 摘要: 利用RT-PCR以及RACE的方法在答氏仿对虾中分离得到1个溶菌酶序列,全长711 bp ,5端非翻译区长度为40\mathrmbp,3端非翻译区为208 bp ,包括加A信号序列AATAAA与多聚A结构,开放阅读框(ORF) 462 bp ,编码153个氨基酸,其中信号肽长度为18个氨基酸,成熟肽长度为135个氨基酸,序列中具有8个半胱氨酸碱基、2个催化反应位点及1个\mathrmCa^2+结合位点,表明该序列属于C型溶菌酶。该序列的获得,有助于为进一步研究溶菌酶的活性。

     

    Abstract: The cDNA of lysozyme was cloned from Parapenaeopsis Hardwickii by RT-PCR and RACE techniques.The complete cDNA sequence of lysozyme is 711 bp,consisting of a 5-\mathrmUTR of 40 bp and 3UTR of 209 bp with a polyadenylation signal sequence AATAAA and a poly (A) tail,the length of ORF is 462 bp encoded a polypeptides of 153 amino acid with a signal peptide of 18 amino acid and a mature protein 135 amino acids.This lysozyme presumed to be a chicken-type lysozyme for it possesses two catalytic sites,eight cysteine residues and one \mathrmCa^2+ binding site.This work provides a basis for further studying of activity of the lysozyme.
