南极海洋微生物Shewanella sp.NJ49的拉曼光谱分析

  • 摘要: 应用激光镊子拉曼光谱技术实时监测、分析南极希瓦氏菌Shewanella sp.NJ49低温降解芳香烃和石油烃过程中单个细胞生长和细胞内外生命物质的动态变化过程,结果表明:NJ49可在低温下有效降解柴油、芳香烃、十六烷烃等烃类物质,可将长直链烃降解为短直链的烷柽;其拉曼光谱均以蛋白质类和葡聚糖类的拉曼信号峰为主要的强峰,其次是脂类物质;拉曼图谱显示NJ49在低温条件下能大量产生β-胡萝卜素,低温下低温微生物产生类胡萝卜素参与调节膜脂肪酸成分的合成以维持细胞膜的流动性,以适应南极地区的低温极端环境。

     

    Abstract: In the present study,laser tweezers Raman spectroscopy (LTRS) was employed to investigate single cell growth and dynamic changes of life materials inside and outside cell of psychrophilic petroleum hydrocarbon-degrading bacterium Shewanella sp.NJ49 isolated from Antarctica.This bacterium can degraded diesel,n-hexadecane and other petroleum hydrocarbons with high efficiency at low temperature;long straight chain hydrocarbons can be decomposed into short straight chain hydrocarbons;The result of Raman spectrum showed that the main signals detected were proteins and carbohydrate signals,and less was lipid signal.The Raman intensity at 1514\mathrm~cm^-1 represented \beta-carotene which involved in the regulation of membrane fluidity in Antarctic bacteria for adaptation in Antactica environment of extreme low temperature.
